Adaptin ear-binding coat-associated protein 2 (NECAP2) is a cytosolic accessory protein involved in the regulation of vesicular trafficking, specifically the endocytic recycling pathway. NECAP2 regulates the recruitment of the clathrin adaptor complex AP-1 to early endosomes, essential for fast recycling of transmembrane proteins such as the epidermal growth factor receptor (EGFR) and transferrin receptor back to the plasma membrane. Loss of NECAP2 impairs this fast recycling route, leading to abnormal endosome morphology and altered receptor trafficking. NECAP2 binds AP-1 via distinct protein-protein interaction interfaces and is considered a pathway-specific regulator of endosomal sorting and vesicle coat formation. It is not currently considered a direct therapeutic target, and there are no known drugs or established safety concerns specifically linked to NECAP2[2][4].
