Adaptor protein complex 2 subunit alpha-1 (AP2A1) is a protein encoded by the AP2A1 gene that forms part of the heterotetrameric AP-2 adaptor complex, crucial for clathrin-mediated endocytosis. The AP-2 complex consists of two large adaptins (alpha: AP2A1/AP2A2, beta: AP2B1), a medium adaptin (AP2M1), and a small adaptin (AP2S1). AP2A1 serves as a cargo adaptor, linking membrane proteins to clathrin to facilitate vesicle formation and receptor internalization. It plays an essential role in regulating cell surface receptor levels, receptor-mediated endocytosis, and synaptic vesicle recycling. AP2A1 recognizes specific endocytic signal motifs and can bind to polyphosphoinositide-containing lipids, positioning the AP-2 complex at the membrane. Dysfunction of the AP-2 complex has been linked to human diseases such as neural tube defects and possibly to neurodegenerative conditions due to its role in autophagic clearance. AP2A1 is not itself a therapeutic target, but its role is essential for membrane trafficking and cellular homeostasis.
