Adaptor-related protein complex 2 subunit beta 1 (AP2B1) is a large subunit of the heterotetrameric AP-2 adaptor complex, which functions as a crucial mediator of clathrin-mediated endocytosis at the plasma membrane[1][2][3][5]. The AP-2 complex acts as a bridge linking clathrin to selected cargo proteins, participating centrally in vesicle formation, cargo selection, and receptor-mediated endocytosis[1][2][3]. AP2B1 specifically contributes to clathrin lattice assembly and provides a scaffold for recruiting endocytic accessory proteins[1][2][3]. Its activity is essential for processes such as synaptic vesicle recycling, and disturbances in AP2B1 or the AP-2 complex have been implicated in neurodevelopmental and neurodegenerative disorders, as well as various pathological states where vesicle trafficking is disrupted[4][5][3]. It is not classified as a therapeutic target, receptor, enzyme, or transporter, but is fundamental for cellular vesicle trafficking and membrane protein sorting[1][2][3].
