Overview

Adaptor-related protein complex 5 subunit beta-1 (AP5B1) is a core component of the AP-5 heterotetrameric adaptor complex, composed of two large subunits (β5—AP5B1 and ζ—AP5Z1), one medium subunit (μ5), and one small subunit (σ5)[1][2][3][4][5]. AP-5 localizes predominantly to late endosomes and mediates trafficking pathways responsible for endosomal protein sorting. Structurally, AP5B1 is similar to the β-adaptin subunits of other AP complexes, containing an N-terminal α-helical solenoid, a flexible linker, and a C-terminal appendage domain[2][3]. AP-5 is evolutionarily ancient and found in many eukaryotes. While AP5B1 itself has not been linked to specific human diseases, the AP-5 complex interacts with proteins (SPG11, SPG15) that, when mutated, cause hereditary spastic paraplegia subtypes[1][3][5]. AP-5 functions in concert with these accessory proteins to facilitate membrane protein cargo sorting; knockout or disruption of AP5B1 in model organisms affects endosomal transport but has not been reported to be embryonic lethal, in contrast to AP-1 and AP-2 complex subunits[2]. No drugs, therapeutic strategies, or clinical biomarkers directly target AP5B1, and it is not considered a current target for pharmacological intervention.
