Overview

Adaptor-related protein complex 5 subunit sigma-1 (AP5S1) is the small subunit of the AP-5 adaptor protein complex, one of five known adaptor protein complexes (AP-1 through AP-5) involved in mediating intracellular membrane trafficking. The AP-5 complex facilitates sorting and transport of cargo proteins within vesicles, particularly in late endosomal compartments[1][2]. AP5S1 does not act as a classical therapeutic target such as a receptor, enzyme, or channel, but is instead a structural and functional protein component of the AP-5 complex. Variants in AP5S1 have not been associated with human disease, but mutations in other AP-5 complex subunits and associated proteins (e.g., AP5Z1, SPG11, ZFYVE26) have been implicated in hereditary spastic paraplegia subtypes[2]. No drugs are currently known to interact with AP5S1, nor is it reported as a biomarker or safety risk.
