Overview

Adaptor-related protein complex 5 subunit zeta-1 (AP5Z1) is a large subunit of the AP-5 adaptor protein complex, which mediates intracellular protein traffic, particularly a sorting step from late endosomes to the Golgi apparatus. AP5Z1 is implicated in endosomal transport, lysosomal homeostasis, and efficient homologous recombination-mediated DNA repair. Damaging variants in AP5Z1 are causative for SPG48, a complicated form of hereditary spastic paraplegia characterized by progressive neurodegeneration and aberrant endolysosomes. AP5Z1 is not generally considered a classical therapeutic target such as a receptor or enzyme, and no drugs are currently known to specifically target this protein[1][2][3][4].
