Overview

Adenosine deaminase domain-containing protein 1 (ADAD1) is a testis-specific RNA-binding protein containing a double-stranded RNA-binding motif and an adenosine deaminase domain; however, its AD domain is catalytically inactive and does not perform RNA editing. Instead, ADAD1 regulates the translation and localization of mRNAs and associated proteins during spermatid development. Loss of ADAD1 causes defective sperm morphology, impaired DNA compaction, abnormal head shaping, reduced motility, and male infertility. The protein is essential for normal differentiation of male germ cells, with roles in nuclear pore complex, LINC complex, manchette structure, and overall regulation of spermiogenesis. It is not currently considered a receptor, enzyme, or therapeutic target, but is crucial for fertility biology.
