Overview

Adenosine deaminase domain-containing protein 2 (ADAD2) is a testis-specific RNA-binding protein characterized by a catalytically inactive adenosine deaminase domain and implicated as a key regulator of male germ cell differentiation. Unlike classical adenosine deaminases, ADAD2 does not exhibit enzymatic activity but is essential for the progression of meiotic spermatocytes to spermatids, with loss-of-function mutations causing maturation arrest and complete male infertility in animal models. ADAD2 is expressed almost exclusively in pachytene spermatocytes within the testis, and its deficiency is associated with sperm development disorders. There are no known therapeutic interventions directed at ADAD2, nor is it implicated in signaling pathways typical of pharmacologic targets[1][3][5].
