Overview

The Adenoviral E1A/E1B replication machinery consists of the early gene products E1A and E1B, which are essential for the adenovirus life cycle and its ability to transform host cells. E1A proteins function as primary transcriptional activators that drive the host cell into the S-phase by binding to the retinoblastoma (pRb) protein, thereby releasing E2F transcription factors (Berk, 2005, PubMed: 16148944). To prevent the host cell from undergoing p53-mediated apoptosis in response to this forced cell cycle entry, the E1B region encodes two key proteins: E1B-55K, which directly binds and inactivates p53, and E1B-19K, a functional homolog of the anti-apoptotic protein Bcl-2 (White, 2001, PubMed: 11483488). This machinery is a major focus in oncology, particularly for the development of oncolytic viruses like H101 and ONYX-015, which utilize E1B-55K deletions to selectively target p53-deficient cancer cells (Bischoff et al., 1996, Science). Beyond virotherapy, these proteins are studied as targets for antiviral drugs designed to inhibit viral replication by disrupting the early regulatory environment established by E1A and E1B.
