Adenovirus antigen-specific cytotoxic T lymphocyte response refers to CD8+ T cells recognizing adenoviral peptides presented by MHC class I on infected cells and executing targeted killing. Activation requires TCR engagement with peptide–MHC I and co-stimulation (e.g., CD28 with CD80/CD86), leading to clonal expansion and acquisition of effector functions. CTLs kill primarily by releasing perforin and granzymes from lytic granules at the immunological synapse and can also induce apoptosis via Fas–Fas ligand interactions, while secreting cytokines such as IFN-γ that inhibit viral replication and enhance antigen presentation. In clinical contexts, strong anti-adenoviral CTL responses limit persistence of adenoviral vectors in gene therapy and are leveraged therapeutically via adoptive transfer of adenovirus-specific CTLs to treat refractory adenoviral infections in immunocompromised patients.
