Overview

The Adenovirus-derived peptide–HLA class I complex is a molecular assembly presented on the surface of cells infected with Adenovirus (Feuchtinger et al., 2004). It comprises a viral antigen fragment, often derived from the conserved Hexon protein, bound to a Human Leukocyte Antigen (HLA) class I molecule (Nelde et al., 2018). This complex is the fundamental unit recognized by the T-cell receptor (TCR) of CD8+ cytotoxic T lymphocytes, triggering an immune response to destroy the infected cell (Leen et al., 2006). In clinical settings, particularly for immunocompromised patients such as those undergoing hematopoietic stem cell transplantation, these complexes are the primary targets for virus-specific T-cell (VST) therapies (AlloVir, 2023). These therapies utilize T cells that are either naturally selected or genetically modified to recognize specific Adenovirus pMHC complexes, providing a targeted approach to control viral replication. Understanding the diversity of these complexes across different HLA alleles is crucial for developing broad-spectrum or personalized immunotherapies (Gerdemann et al., 2012).
