Overview

The Adenovirus penton protein peptide-HLA class I complex is a molecular assembly consisting of a short peptide fragment derived from the adenovirus penton base protein bound within the groove of a Human Leukocyte Antigen (HLA) class I molecule (PMID: 15140913). This complex is expressed on the surface of cells infected with adenovirus, serving as a critical signal for the immune system to identify and eliminate the virus (PMID: 21835945). CD8+ cytotoxic T lymphocytes (CTLs) recognize these specific pMHC complexes via their T-cell receptors (TCRs), triggering an effector response that includes the release of perforins and granzymes to induce apoptosis in the target cell (UniProt P03276). In clinical settings, these complexes are primary targets for adoptive immunotherapies, such as virus-specific T-cells (VSTs), which are used to treat life-threatening adenovirus infections in immunocompromised patients, particularly those undergoing hematopoietic stem cell transplantation (PMID: 24501210). Understanding the specific epitopes within the penton protein and their binding affinity to various HLA alleles is essential for developing broad-spectrum or personalized antiviral therapies. While the hexon protein is often the dominant source of T-cell epitopes, the penton protein also contains highly conserved sequences that are vital for generating a robust and multi-antigenic immune response (PMID: 18061611).
