Adenylate cyclase (AC) is an enzyme that catalyzes the conversion of ATP to cyclic AMP (cAMP) and pyrophosphate. It plays a central role in signal transduction by producing cAMP, a second messenger that regulates various downstream targets. There are ten membrane-bound isoforms (AC1-AC10) and one soluble isoform (sAC) in mammals, each with unique regulatory properties and tissue distributions. AC activity is regulated by G protein-coupled receptors, calcium, and other factors. Defects in AC signaling have been implicated in various diseases. Although many drugs target upstream GPCRs, direct modulators of AC isoforms are under investigation.
