Overview

Adenylate cyclases are a family of enzymes that synthesize cAMP from ATP in response to extracellular signals, integrating transmembrane and cytosolic domains. Each isoform (ADCY1, ADCY2, ADCY5) has a distinct tissue distribution and regulatory profile: ADCY1 is prominent in the brain and involved in memory and learning; ADCY2 is found in many tissues and regulated by calmodulin-insensitive mechanisms via G-protein beta-gamma subunits; ADCY5 is highly expressed in the heart and testis and plays roles in cardiac and movement control. They are key regulatory nodes in cell signaling, making them therapeutic targets for cancer, cardiovascular, neurological, and metabolic diseases. Drugs that increase or inhibit their activity (such as forskolin and various purine analog inhibitors) alter cAMP-dependent pathways for experimental or therapeutic purposes. Mutations and altered expression of ADCY isoforms are associated with pathological states and may serve as biomarkers or pharmacogenetic risk factors.
