Overview

Adenylate cyclase type 10 (ADCY10) encodes a soluble adenylyl cyclase, a cytoplasmic enzyme responsible for catalyzing the conversion of ATP to cyclic AMP (cAMP), a ubiquitous second messenger involved in intracellular signaling. Unlike transmembrane adenylyl cyclases, ADCY10 is regulated by intracellular bicarbonate and is not controlled by G protein or forskolin. It plays critical roles in sperm function (including maturation, motility, and capacitation), pH regulation in renal and epididymal tissues, ciliary movement, and localized cAMP signaling within various cellular compartments (cytosol, nucleus, mitochondria). Mutations in ADCY10 are associated with male infertility and absorptive hypercalciuria. There are currently no approved therapeutic agents targeting ADCY10, but research continues into its physiological relevance and potential as a drug target.
