Overview

Adenylate cyclase type 3 (ADCY3) is a membrane-associated enzyme that catalyzes the conversion of ATP to cyclic adenosine monophosphate (cAMP) in response to G-protein signaling[3]. It is widely expressed in human tissues, especially in primary neuronal cilia and olfactory neuroepithelium, and plays a central role in the regulation of cellular signal transduction, energy metabolism, glucose homeostasis, and neuroendocrine functions[1][3][4]. Genetic variants in ADCY3 are linked to predisposition to obesity, type 2 diabetes, and potentially neuropsychiatric conditions such as major depressive disorder. Recent studies implicate ADCY3 in cancer biology, notably as a tumorigenic factor in gastric cancer via the cAMP/PKA/CREB pathway[2]. Currently, there are no approved drugs that specifically or directly target ADCY3 in clinical use.
