Adenylate cyclase type 8 (ADCY8) is a membrane-bound enzyme that catalyzes the conversion of ATP to cyclic AMP (cAMP) in response to increases in intracellular calcium, integrating Ca²⁺ and cAMP signaling. ADCY8 is highly expressed in neurons, specifically at synaptic sites, and plays a key role in synaptic plasticity, learning, memory, and modulation of neuronal signaling. In the pancreas, ADCY8 is a critical component for glucose-stimulated cAMP production and insulin secretion. ADCY8 is regulated by G protein-coupled receptors and Ca²⁺/calmodulin, and its dysfunction has been linked to neurological and metabolic diseases, including anxiety, dissociative amnesia, and impaired insulin secretion in diabetes.
Drugs targeting the cAMP pathway may indirectly modulate ADCY8 activity by affecting upstream (G protein-coupled receptors) or downstream (protein kinase A, CREB) signaling
