Adenylate kinase 8 (AK8) is a cytosolic enzyme belonging to the adenylate kinase family that catalyzes the reversible transfer of phosphate groups between nucleoside triphosphates and nucleoside monophosphates, playing a key role in maintaining cellular energy homeostasis. AK8 is unique among human adenylate kinases, possessing two complete and active kinase domains in its polypeptide chain. Its activity is highest toward AMP, but it can also phosphorylate other nucleotides such as dAMP, CMP, and dCMP, using ATP or GTP as phosphate donors. AK8 is predominantly expressed in tissues with motile cilia and is associated with some rare genetic disorders, such as primary ciliary dyskinesia and reticular dysgenesis. There is currently no evidence that AK8 is a direct therapeutic target or that drugs interact with it, and it is not currently used as a biomarker or associated with specific safety concerns.
