Overview

Adenylate kinase 9 (AK9) is a member of the adenylate kinase enzyme family and functions as a broad-specificity nucleoside phosphate kinase. It catalyzes the interconversion of nucleoside monophosphates, diphosphates, and triphosphates—particularly involving AMP, dAMP, CMP, and dCMP—thus maintaining nucleotide and energy homeostasis within the cell. This activity is important both for basal cell metabolism and for activating nucleoside analogs used in some antiviral and cancer therapies. Mutations in AK9 have been directly associated with rare cases of congenital myasthenic syndrome, where it likely supports N-glycosylation at the neuromuscular junction, and altered AK9 activity has been observed in certain cancers such as malignant melanoma, suggesting a broader role in disease. As an intracellular metabolic regulator, AK9 is emerging both as a candidate drug target and as a disease modifier in nucleotide imbalance disorders.
