Overview

Adenylate kinase isoenzyme 6 (AK6) is a broad-specificity nucleoside monophosphate kinase that catalyzes the reversible transfer of the terminal phosphate group between nucleoside triphosphates and monophosphates, showing both adenylate kinase and ATPase activity[1][2][5]. AK6 is predominantly localized to the nucleus, where it helps regulate nuclear nucleotide pools and energy homeostasis[1][2]. Unlike other adenylate kinases, AK6 can use all nucleoside triphosphates (NTPs and dNTPs) as phosphate donors and has unique sequence and structural features[1][3][5]. It is implicated in ribosome biogenesis, specifically in the 18S rRNA maturation step, via its ATPase activity, and may also play a role in Cajal body formation[2][5]. AK6 is considered an enzyme target, but currently there are no reported drugs known to specifically target it, nor established patient selection biomarkers or recorded safety concerns. Disease associations are limited, with reticular dysgenesis being noted in gene-disease databases[2].
