Adenylosuccinate synthetase (EC 6.3.4.4) is a key enzyme in purine metabolism, catalyzing the first committed step in the de novo biosynthesis of adenosine monophosphate (AMP). It facilitates the guanosine triphosphate (GTP)-dependent conversion of inosine monophosphate (IMP) and L-aspartic acid to N(6)-(1,2-dicarboxyethyl)-AMP (adenylosuccinate), guanosine diphosphate (GDP), and phosphate. It plays a central role in both the de novo and salvage pathways of purine nucleotide biosynthesis. Enzyme activity is competitively inhibited by AMP and IMP.
