Enterococcus faecalis Ace adhesin (Adhesin to collagen of E. faecalis) is a cell-wall-anchored surface protein belonging to the Microbial Surface Component Recognizing Adhesive Matrix Molecules (MSCRAMM) family. It plays a critical role in the pathogenesis of E. faecalis infections by mediating the attachment of the bacteria to host extracellular matrix (ECM) components, specifically collagen types I and IV and laminin. This adherence is a vital initial step in the development of infective endocarditis and urinary tract infections. The expression of Ace is conditionally regulated and significantly upregulated in the presence of host factors such as serum, bile salts, or elevated temperatures. Due to its essential role in host colonization and its presence in clinical isolates, Ace is considered a promising therapeutic target for the development of vaccines and anti-adhesion therapies. Experimental studies have shown that active immunization with recombinant Ace or passive administration of anti-Ace antibodies can provide protection against experimental endocarditis in animal models.
