Adhesion G protein-coupled receptor A3 (ADGRA3), previously known as GPR125, is a member of the adhesion class of G protein-coupled receptors (GPCRs) characterized by long N-terminal extracellular regions with protein-protein interaction motifs such as Ig-like domains and leucine-rich repeats. ADGRA3 is classified as an orphan receptor since no endogenous ligands have been identified to date. It is highly expressed in the testis and prostate, with evidence suggesting a role in the development of spermatogonia and germ cell function. The receptor has also been implicated as a marker for adult stem and progenitor cells, and its expression is upregulated following brain injury, suggesting possible wider roles in tissue homeostasis and repair. However, direct biological functions and disease associations remain only partially defined, and it is not yet a target for approved therapeutics[1].
