Adhesion G protein-coupled receptor B2 (ADGRB2, also known as Brain-specific angiogenesis inhibitor 2 or BAI2) is a member of the adhesion GPCR subfamily B, characterized structurally by a large extracellular region containing multiple adhesion motifs and a seven-transmembrane (7TM) domain[3][6][5]. It functions as an orphan GPCR involved in mediating cell-cell adhesion and likely plays roles in neural development, synaptic formation, angiogenesis inhibition, and signal transduction through the NFAT pathway[1][5][6][9]. ADGRB2 is primarily expressed in the brain and participates in diverse physiological and pathological processes, including neurodevelopment, psychiatric disorders, angiogenesis regulation (especially in response to ischemic events), and perhaps tumorigenesis[2][7][3]. While it possesses clear therapeutic potential as a receptor, no specific drugs or approved clinical therapies currently target ADGRB2, although it remains a candidate for future pharmacological intervention within the adhesion GPCR family[8][10].
