Overview

Adhesion G protein-coupled receptor E1 (ADGRE1), also known as EMR1, is a member of the adhesion GPCR family characterized by an extended extracellular region containing several EGF-like domains and a seven-transmembrane helix region, linked via a GAIN domain[2][3][4]. It is predominantly expressed on cells of the myeloid lineage: in humans, notably on eosinophils (and minimally on macrophages); in mice, its homolog F4/80 is a widely used marker for tissue-resident macrophages[3][4]. ADGRE1 is involved in cell adhesion, migration, and intercellular signaling, contributing to immune responses, inflammation, and tissue homeostasis[1][2][3][4]. It has roles in mediating macrophage activation and inflammatory signaling, particularly in contexts like metabolic inflammation and chronic immune disorders[1][2]. The receptor is a target for experimental therapies aiming at depleting eosinophils in certain inflammatory and allergic diseases using afucosylated antibodies[3].
