Adhesion G protein-coupled receptor E3 (ADGRE3, also known as EMR3) is a member of the adhesion GPCR family predominantly expressed in human myeloid cells, such as monocytes, macrophages, myeloid dendritic cells, and mature granulocytes[1][2][7][8]. Structurally, ADGRE3 features an extended extracellular region with epidermal growth factor (EGF)-like domains linked to a seven-transmembrane (TM7) domain via a mucin-like spacer[1][2][7]. It is an orphan receptor—its endogenous ligand is unknown, although surface ligands are suggested to be present on macrophages and neutrophils[1][6][7]. ADGRE3 is implicated in the regulation of immune and inflammatory responses, likely mediating cell-cell interactions among myeloid cells. Its expression is upregulated in inflammation and some cancers, hinting at roles in disease pathogenesis, but it is not yet a target for approved drugs or therapeutics[1][2][4][7]. There are no mouse or rat orthologues of this receptor[6].
