Overview

Adhesion G protein-coupled receptor G7 (ADGRG7) is a member of the adhesion class of G protein-coupled receptors (GPCRs), characterized by a long extracellular N-terminal region often containing protein modules and linked to the 7-transmembrane domain via the GPCR-Autoproteolysis INducing (GAIN) domain[2][3]. ADGRG7 is considered an orphan receptor, as specific endogenous ligands have not yet been identified[3][4]. Its physiological roles are being elucidated but currently include potential involvement in regulating body weight, gastrointestinal motility, skeletal development, and immune system signaling[1][2]. Clinical evidence links ADGRG7 to oncogenic processes such as gene fusions in T-cell lymphoblastic lymphoma, and its expression has been correlated with cancer progression and metastasis in several tumor types[1][4]. ADGRG7 expression has also been implicated in certain skeletal diseases, and a peptide derived from ADGRG7 can trigger a T-cell immune response, suggesting a possible role in immune recognition[1]. Currently, there are no known drugs that directly target ADGRG7, nor defined mechanisms of action for modulation, and it remains under investigation as both a functional and clinical molecule.
