Adhesion molecule with Ig-like domain 3 (AMIGO3) is a membrane protein of the immunoglobulin superfamily, containing leucine-rich repeat motifs and Ig-like domains, widely expressed in astrocytes, oligodendrocytes, and neurons. Its main biological functions are mediating heterophilic cell-cell adhesion, modulating synaptic plasticity, and inhibiting axon regeneration and myelin production after CNS damage via the ROCK/RhoA signaling pathway. AMIGO3 is upregulated after neural injury and is considered a therapeutic target for demyelinating conditions and abnormal neural circuitry in diseases such as epilepsy and spinal cord injury. Antagonism of AMIGO3 has shown promise in preclinical models for improving myelin repair and neural regeneration.
Inhibition of AMIGO3 can promote myelin regeneration and axon outgrowth, likely via ROCK/RhoA pathway modulation. It has potential to substitute for LINGO1 in the Nogo receptor complex, mediating myelin-induced axon outgrowth inhibition.
