The adipocyte plasma membrane lipid bilayer is the fundamental structural boundary of the fat cell, composed of a complex arrangement of phospholipids, sphingolipids, and cholesterol (PubMed: 25650186). It serves as a critical platform for metabolic signaling, housing essential receptors and transporters such as the insulin receptor and GLUT4, which are vital for glucose homeostasis (NIH: PMC3518445). In pathological states such as obesity and type 2 diabetes, the lipid composition of this bilayer often shifts toward increased saturation, leading to decreased membrane fluidity and impaired signal transduction, a state closely linked to insulin resistance (PubMed: 22964371). While not a traditional protein target, the physical properties of the membrane are the focus of membrane-lipid therapy (melitherapy), which utilizes lipophilic compounds like BGP-15 to restore normal cellular function by altering membrane organization (Escribá et al., 2015). However, because it is a broad cellular structure rather than a discrete molecular entity, it is generally not classified as a specific therapeutic target in conventional drug discovery.
