The adipocyte plasma membrane phospholipid bilayer is the fundamental structural boundary of the fat cell, consisting of a semi-permeable arrangement of amphipathic lipids that separates the intracellular environment from the extracellular matrix (Lodish et al., 2000). It serves as a critical regulatory hub for lipid metabolism and systemic energy homeostasis, hosting essential proteins such as GLUT4 and various hormone receptors (Saltiel and Kahn, 2001). In therapeutic applications, this bilayer is the direct target of cytolytic agents like deoxycholic acid, which act as detergents to physically disrupt the phospholipid arrangement (FDA, 2015). This disruption leads to the loss of membrane integrity, resulting in adipocytolysis or the permanent destruction of the fat cell (Rotunda et al., 2004). This mechanism is utilized in aesthetic medicine for the reduction of submental fat, where the targeted lysis of adipocytes triggers a localized inflammatory response and subsequent tissue remodeling (Dayan et al., 2016). However, the non-selective nature of membrane disruption poses risks to surrounding tissues, necessitating precise delivery to avoid damage to nerves or skin (Kythera Biopharmaceuticals, 2015).
