The adipogenic transcriptional program is a highly orchestrated cascade of gene expression events that transforms undifferentiated mesenchymal stem cells into mature, lipid-storing adipocytes (Rosen & MacDougald, 2006, Nature Reviews Molecular Cell Biology). This process is primarily driven by a core set of transcription factors, most notably Peroxisome Proliferator-Activated Receptor gamma (PPAR-gamma) and members of the CCAAT/enhancer-binding protein (C/EBP) family (Farmer, 2006, Genes & Development). These factors work in concert to activate the expression of thousands of genes involved in insulin sensitivity, lipid synthesis, and adipokine secretion (Lefterova & Lazar, 2009, Trends in Endocrinology & Metabolism). Dysregulation of this program is central to the development of metabolic disorders such as obesity and type 2 diabetes (Evans et al., 2004, Nature Medicine). While the program itself is a complex biological process rather than a single protein, its key components—particularly PPAR-gamma—have been successfully targeted by drugs like thiazolidinediones to improve metabolic health (Yki-Järvinen, 2004, New England Journal of Medicine).
