Adipogenin (ADIG), also known as Small adipocyte factor 1 (SMAF1), is a small (~10 kDa) adipocyte-enriched transmembrane protein. It is predominantly expressed in adipocytes and is induced during adipocyte differentiation (adipogenesis). ADIG appears to regulate adipogenesis and adipocyte function, especially by modulating leptin secretion—it is required for normal formation of adipose tissue and maintenance of fat mass. Loss of ADIG in mouse models impairs adipogenesis, reduces fat mass accumulation (especially under high-fat diet conditions), and decreases leptin secretion from adipose tissue. Variants near the human ADIG gene have been linked to leptin levels via genome-wide association studies, suggesting a potential genetic influence on energy balance and obesity susceptibility. No evidence currently supports Adipogenin as a receptor, enzyme, transporter, or transcription factor, and it is not a known drug target or clinical biomarker. Its primary scientific interest is in adipose biology and metabolic regulation.
