Overview

Adjuvant-mediated immune system stimulation refers to the process by which adjuvants, substances included in vaccines and certain immunotherapies, enhance the body's immune response to an antigen. Adjuvants achieve this by activating innate immune receptors (such as TLRs and NLRs) on antigen-presenting cells, increasing antigen uptake and presentation, upregulating co-stimulatory molecules and cytokine release, and promoting polarization toward either antibody or cellular immune responses. This enhanced priming leads to stronger and longer-lasting adaptive immunity, improved protection against infection, and is increasingly essential in modern vaccines utilizing weakly immunogenic antigens, as well as for cancer immunotherapy. Adjuvant stimulation is a process, not a discrete molecular target, and involves a spectrum of molecular pathways and cellular actors, with both established and emerging adjuvants varying in efficacy and safety
