Overview

ADP-ribosylation factor-like GTPase 17B (ARL17B) is a member of the ARF (ADP-ribosylation factor) family of small GTPases, specifically the ARF-like (ARL) group within the Ras superfamily. It is predicted to possess GTP binding and intrinsic GTPase activity, suggesting a regulatory function in intracellular protein and vesicle transport, potentially modulating vesicle budding and uncoating at the Golgi apparatus. ARL17B localizes to the Golgi apparatus and is implicated in membrane trafficking, with genetic, epigenetic, and evolutionary studies pointing to a role in disorders of the central nervous system, such as progressive supranuclear palsy, Parkinson’s disease, and developmental dyslexia—often due to its altered expression or involvement in chromosomal rearrangements in the 17q21.31 region. The exact molecular and cellular function of ARL17B remains to be fully elucidated, and to date, no drugs are known to target this protein, nor is it regarded as a validated pharmacological target.
