Overview

ADP-ribosylation factor-like protein 11 (ARL11) is a member of the Ras superfamily of small GTPases related to the ADP-ribosylation factor family. It functions as a tumor suppressor, integrating pro-apoptotic and immune-regulatory signals. ARL11 induces caspase-dependent apoptosis and suppresses tumor formation in various cancers, including lung, prostate, ovarian, breast, and hematological malignancies. It also regulates macrophage activation by positively influencing ERK1/2 MAPK signaling in response to inflammatory stimuli such as LPS, thus connecting innate immune activation with cell death pathways. Certain genetic variants in ARL11 are associated with increased familial cancer risk and may serve as potential biomarkers for susceptibility or disease prognosis. No known drugs directly target ARL11, but its key regulatory role in apoptosis and immune signaling positions it as a potential therapeutic and diagnostic target in oncology and immunology[1][2][3].
