ADP-ribosylation factor-like protein 15 (ARL15) is a member of the ARF family of small GTPases, structurally related to ADP-ribosylation factors but with divergent functions. ARL15 is ubiquitously expressed in human tissues and localizes predominantly to the Golgi apparatus, with additional presence at the plasma membrane and in vesicular compartments. Its key roles include regulation of adipocyte differentiation and adiponectin secretion, mediating intracellular trafficking processes, and acting as a positive regulator of TGFβ family signaling via direct interaction with Smad4, thereby influencing Smad-complex assembly and signaling. Genetic studies have associated ARL15 variants with metabolic syndromes, including insulin resistance, obesity, type 2 diabetes, dyslipidemia, and lipodystrophy, as well as rheumatoid arthritis. Although no approved therapeutic drugs directly target ARL15, its fundamental participation in these pathways make it of significant interest as a potential therapeutic target for metabolic and inflammatory diseases[1][2][3][5].
