Overview

ADP-ribosylation factor-like protein 5A (ARL5A) is a member of the ARF family of small GTP-binding proteins, structurally related to ADP-ribosylation factors but lacking their enzymatic activity. ARL5A is primarily localized to the nucleus/nucleolus, interacts with HP1alpha, and is developmentally regulated. It may play a role in nuclear and nucleolar dynamics or signaling cascades during embryonic development, although its precise cellular actions remain incompletely defined. Unlike canonical ARF proteins, ARL5A lacks ADP-ribosylation activity and its direct involvement in signaling, trafficking, or disease mechanisms is still under investigation. Disease associations include mucinous stomach adenocarcinoma and Dyggve-Melchior-Clausen disease, but no drugs or pharmacological modulators are currently reported for this target[3][6][7].
