Overview

ADP-ribosylhydrolase like 1 (ADPRHL1) is a pseudoenzyme within the ADP-ribosylhydrolase protein family, strongly expressed in the developing heart of vertebrates, particularly in myocardial cells involved in chamber growth[1][2]. Unlike active family members, ADPRHL1 lacks the amino acid residues necessary for ADP-ribosylhydrolase catalytic activity and does not show enzymatic function[1][2]. Its protein is essential for proper assembly of cardiac myofibrils and heart chamber outgrowth; deficiency in ADPRHL1 disrupts myofibril structure, impairs actin filament organization, and prevents normal ventricle development[1][2]. ADPRHL1 localizes to structures close to the sarcomeric Z-disc, implicating it in actin dynamics and myofibril formation[1][2]. While not a direct drug or therapeutic target and not associated with any known interacting drugs or clinical biomarkers, mutations or deficiencies in ADPRHL1 are linked to severe defects in cardiac development, underscoring its specialized role in heart morphogenesis rather than in classical metabolism or signaling pathways typical of therapeutically targeted enzymes[1][2][3].
