ADP-ribosyltransferase 5 (ART5) is a member of the arginine-specific ADP-ribosyltransferase family, catalyzing the transfer of ADP-ribose from NAD+ onto arginine residues in target proteins, thereby regulating their function. Unlike membrane-anchored family members ART1 and ART2, ART5 is secreted and exhibits unique catalytic properties, such as dual catalytic activity dependent on NAD+ concentration and auto-regulatory modification through auto-ADP-ribosylation. These features contribute to ART5’s distinct biology and tissue specificity. ART5 has been implicated in cell signaling and protein regulation and has emerging relevance as a biomarker in cancer research.
