ADPGK antisense RNA 1 (ADPGK-AS1) is a long non-coding RNA located on chromosome 1q21 that does not code for a protein but plays a regulatory role in gene expression and cell phenotype[1][2][4][5][6]. It is expressed in various tissues, notably in immune cells such as macrophages, where it regulates mitochondrial metabolism and influences the macrophage phenotype (M1 pro-inflammatory versus M2 tumor-promoting), especially in the tumor microenvironment[1][4]. Elevated expression of ADPGK-AS1 is linked with increased tumor cell proliferation, resistance to apoptosis, migration, and poor clinical prognosis in several cancers, including lung, breast, cervical, osteosarcoma, and hepatocellular carcinoma[2][4][5][6]. Its function appears to be especially important in shaping the metabolic and immunological landscape of tumor-associated macrophages, thereby influencing tumor growth and response to therapy. Silencing ADPGK-AS1 in tumor-associated macrophages reverses the tumor-promoting phenotype and suppresses tumor growth in preclinical models, suggesting that ADPGK-AS1 may serve as a candidate for therapeutic targeting in cancer[1][4]. However, there are no known drugs or direct therapeutic agents clinically targeting ADPGK-AS1 as of the current research.
