Adrenomedullin (ADM) is a multifunctional 52-amino acid peptide hormone structurally related to the calcitonin gene-related peptide (CGRP) family. It is widely expressed throughout the body and plays a critical role in regulating vascular tone, blood pressure, and cardiovascular protection. ADM exhibits anti-inflammatory effects, maintains vascular integrity, and promotes angiogenesis and tissue repair. It primarily binds to receptors composed of the Calcitonin-like receptor (CLR) and Receptor activity-modifying proteins (RAMPs), activating downstream signaling pathways that mediate its diverse biological functions. Elevated plasma levels are observed in various cardiovascular diseases. Potential therapeutic applications include treatment/prevention of cardiovascular disorders, inflammatory diseases, and tissue injuries.
Adrenomedullin exerts its effects primarily through binding to receptors composed of Calcitonin-like receptor (CLR) and Receptor activity-modifying proteins (RAMPs). Upon ligand binding, key downstream pathways include cAMP production → PKA activation, PI3K/Akt pathway activation, and ERK phosphorylation. These lead to reduced myosin light chain phosphorylation, increased cortical actin formation, and inhibition of pro-inflammatory responses.
