Overview

SSX2IP (Afadin- and alpha-actinin-binding protein) is an adaptor protein implicated in the organization of adherens junctions and centrosome maturation within the cell. It binds to afadin and alpha-actinin, connecting the nectin-afadin system with the E-cadherin–catenin complex, thereby helping to organize the cytoskeleton and maintain cell-to-cell junction integrity. SSX2IP also interacts with WDR8, playing essential roles in centrosome maturation, spindle dynamics, and ciliogenesis, all functions relevant to normal cell division, cell migration, and tissue organization. Its association with cancer-testis antigens (such as SSX2) suggests possible cancer relevance, particularly in synovial sarcoma and leukemia. Although not yet an established therapeutic target, its molecular and functional roles position it as a candidate for further investigation in cell cycle control and malignancy.
