AFDN divergent transcript (AFDN-DT) is a long non-coding RNA gene situated upstream of the Afadin (AFDN) locus on chromosome 6. It is mainly located in the nucleus and exerts its tumor-suppressive effects by binding directly to chromatin and regulating the transcription of genes critical for cell proliferation, metastasis, and transformation, particularly in gastric cancer. This regulatory function involves interaction with transcription factor motifs and direct binding to promoters of important cancer-related genes (such as JUN, FOS, and EGR1). Repression of AFDN-DT correlates with enhanced cancer aggressiveness, while restoration of its expression (e.g., by DNA methylation inhibitors) has been linked to growth inhibition of cancer cells[1][5].
