Afferent nerve fiber activation refers broadly to the initiation of action potentials in sensory neurons that carry information from peripheral tissues toward the central nervous system. These primary afferents include myelinated Aβ and Aδ fibers—which transmit touch/pressure/pain—and unmyelinated C fibers—which are primarily responsible for slow pain transmission and neurogenic inflammation responses. The type of stimulus (mechanical, thermal, chemical) determines which subset(s) are activated. This physiological event underlies all somatosensory signaling including touch perception and nociception—the neural processes encoding noxious stimuli leading to pain sensation. While modulation or inhibition of these processes is therapeutically relevant for analgesia or anesthesia, "afferent nerve fiber activation" itself does not refer to any one molecule but encompasses diverse cellular mechanisms across multiple cell types.
