Overview

The afferent neural pathway between the stomach and central nervous system refers to the network of sensory neurons—primarily those traveling via the vagus nerve—that transmit information about mechanical and chemical conditions in the stomach to the brainstem and higher centers. This forms a key part of the gut-brain axis, which enables bidirectional communication between the gastrointestinal tract and central nervous system. The majority (~80%) of fibers in the vagus nerve are afferent, carrying signals from visceral organs including the stomach up to nuclei in the medulla oblongata such as the nucleus tractus solitarius. These pathways play crucial roles in regulating digestion, satiety/appetite signaling, immune responses through inflammatory reflexes, and overall homeostasis. While these pathways are essential for normal physiology—and their dysfunction is implicated in various diseases—they do not constitute a single molecular therapeutic target but rather represent complex circuits involving multiple cell types and neurotransmitters.
