Overview

Aftiphilin is a clathrin-associated adaptor protein that plays a significant role in intracellular membrane trafficking and protein sorting (UniProt Q6ULP2). It primarily functions at the trans-Golgi network and endosomes, where it interacts with the AP-1 adaptor complex and clathrin to facilitate vesicle formation and cargo transport (Hirst et al., 2005, Molecular Biology of the Cell). In neurons, Aftiphilin is localized to synaptic vesicles and is thought to participate in the recycling of these vesicles, thereby influencing neurotransmission (Burman et al., 2005, FEBS Letters). Beyond its role in the nervous system, it is involved in the regulated exocytosis of Weibel-Palade bodies in endothelial cells, which is crucial for blood clotting (Lui-Roberts et al., 2008, Journal of Cell Science). While Aftiphilin is not currently a target for any FDA-approved drugs, its dysregulation has been observed in various cancers and neurological conditions, making it a subject of interest for future therapeutic research (NCBI Gene ID: 54812). The protein's broad involvement in essential trafficking pathways suggests that any future drug development would need to carefully manage potential off-target effects on cellular homeostasis.
