Overview

AHNAK nucleoprotein is an exceptionally large (~700 kDa) scaffold protein comprised of N-terminal (containing a PDZ domain), central repeated units, and a C-terminal domain rich in interaction and regulatory sites[3]. It is ubiquitously expressed and localizes variably in the nucleus, cytoplasm, cell membrane, and other compartments depending on cell type and conditions. AHNAK interacts with L-type calcium channel subunits and actin, organizing subsarcolemmal cytoskeletal networks and modulating calcium influx and membrane repair. Its molecular structure features a series of highly conserved repeat segments predicted to fold into propeller-like β-strand domains, positioning it as a key organizer of cellular architecture and signaling machinery. AHNAK is connected to diverse physiological and pathological roles, especially in regulating calcium signaling, cell migration, blood-brain barrier formation, and cancer progression. It does not currently serve as a direct therapeutic target but is a significant prognostic biomarker in oncology and cardiovascular research[1][2][3][4].
