Akirin-2 (AKIRIN2) is a small nuclear regulatory protein that acts as a molecular adapter, bridging multi-protein complexes and playing key roles in embryonic development, innate and adaptive immune responses, myogenesis, and brain development. It is essential for the nuclear import of 20S proteasomes in vertebrates by binding both proteasomes and nuclear import receptors, enabling nuclear protein quality control and degradation. AKIRIN2 regulates chromatin remodeling and transcription, particularly at immune gene promoters like IL-6, by interacting with chromatin remodeling complexes (e.g., BAF/SWI-SNF) and adaptor proteins (e.g., IκB-ζ). Loss or knockdown of Akirin-2 impairs the expression of inflammatory cytokines, cell cycle regulators, and muscle differentiation genes, and is associated with defective proliferation, survival, and tissue-specific developmental processes. Although not a direct therapeutic target, its pivotal role in gene regulation and protein homeostasis links it to various disease processes, particularly inflammation, immunity, and cancer.
