Overview

The Akt-mTOR-S6K signaling axis is a central intracellular pathway that regulates key cellular processes such as cell growth, proliferation, survival, metabolism, and protein synthesis. This axis integrates signals from nutrients, growth factors, and cellular energy status to coordinate anabolic and catabolic responses in the cell. It involves Akt, mTOR (in mTORC1 and mTORC2 complexes), and S6 Kinases, each with specific roles and feedback mechanisms. Hyperactivation is common in many cancers, while dysregulation is also implicated in neurological disorders. Targeting this axis has been a major focus for drug development, but is limited by compensatory feedback and side effects.
