Target intelligence / Profile preview
Albumin receptors and albumin-mediated uptake mechanisms encompass a variety of proteins that facilitate the transport, recycling, and cellular internalization of serum albumin. The neonatal Fc receptor (FcRn) is a primary regulator of albumin homeostasis, rescuing it from lysosomal degradation through a pH-dependent recycling pathway, which accounts for its exceptionally long half-life (Bern et al., 2015, Journal of Controlled Release). In the vascular endothelium, glycoprotein 60 (gp60 or albondin) mediates the transcytosis of albumin into the interstitial space via caveolae-mediated endocytosis (Schnitzer, 1992, American Journal of Physiology). Furthermore, the matricellular protein SPARC (Secreted Protein Acidic and Rich in Cysteine) is often overexpressed in tumors, where it binds and sequesters albumin, facilitating the accumulation of albumin-bound drugs like nab-paclitaxel (Abraxane) (Ji et al., 2024, Frontiers in Pharmacology). Renal reabsorption of albumin is primarily handled by the megalin-cubilin complex in the proximal tubules, preventing protein loss in the urine (Birn & Christensen, 2006, Kidney International). These mechanisms are strategically exploited in drug development to enhance the pharmacokinetics of therapeutic proteins and to achieve targeted delivery in oncology. However, therapeutic modulation of these pathways, particularly FcRn, requires careful management to avoid adverse effects like hypoalbuminemia and systemic edema.
Drugs utilize these mechanisms for enhanced half-life via FcRn-mediated recycling, targeted tumor delivery via gp60-mediated transcytosis and SPARC-mediated sequestration, or therapeutic inhibition of FcRn to deplete pathogenic antibodies.
